Bosses have invested £250,000 in the new store.
Poundworld and Discount UK trading director Chris Edwards Junior said: "We're really delighted to be opening our first store in Stafford and to be creating 40 new jobs for the town.
"By combining a Poundworld and Discount UK store in one outlet, we are offering our customers the best possible value and allowing them to take advantage of great offers on higher-value items over £1 that we aren't able to sell in a purely Poundworld store. We look forward to welcoming customers."
Poundworld was founded in 1997 and has more than 140 stores across the UK. Directors are aiming to open a further 35 shops by the end of 2012. There are more than 30 Discount UK stores nationwide but only a handful of outlets featuring both brands.
